The Influence of Postnatal Exposure to Intensive Sound on the Auditory System in Rat

Newborn SD rats were divided into three groups:one group for control and two for experiments. Young rats were exposed to high-intensive pure-tone (4kHz,80db spl)for 12 h daily from postnatal days 1 and 9. The changes of succinic dehydrogenase activity of the hair cells of basilar membrane were studied at about 90 days after birth.We found that two experimental groups succinic dehydrogenase activity of the hair cells of middle turn basilar membrane decreased when compared with that of control group, especially the outer hair cells.The histochemical changes of the succinic dehydrogenase activity suggest that the disturbance in energy metabolism is closely associated with postnatal exposure of high-intensive pure-tone.While the single unit responses to pure tone stimuli at dorsal cochlear nucleus (DCN) neurons were studied electrophysi-ologically,the distribution of best frequency (BF)of units from postnatal days 1 and 9, when compared with the control group,showed an abnormal lossing centered around the region of the audiogrom occupied by the tones to which the annimals had been exposed. The results suggested that postnatal exposure of high-intensive pure-tone directly damaged the corresponding region of the basilar membrane of the cochlear,and dereased pro-jection to the higher central auditory nucleus.As a result,units of the corresponding re-gion of the dorsal cochlear nucleus were missing. The result of this experiment indicates that the damage of the auditory system is getting more serious with earlier exposure.

To study the morphological and quantitative changes of mast cell (MC) in scalding wound healing,the full-thickness scalding model were established in rats.The HE and Toluidine blue stain were used to detect the changes of MC in morphology and quantity, granulation tissue, and collagen fibers.The results showed that MC was degranulated in different degree after scalding. The quantity of MC decreased 24 hours later and reached the lowest level on the 3rd day. Then the quantity of MC increased gradually and reached the highest level on the 28th day. After that the quantity of MC decreased again.A little granulation tissue grew in the bottom and edge of the wound at 3 days after scalding. On the 14th day,the collagen fibers were found in the area of granulation tissue formed in the earlier stage.These findings suggest that there are three stages in the quantitative change of MC after scalding in rats,and the development of granulation tissue and collagen fibers is correlated with the changes of MC in the wound healing.

Using the three times (interval 4 hours) continual partial hepatectomy (CPH) rat as model,the changes in activity of the acid phosphatase (ACP), alkaline phosphatase (AKP),and content of constitutive heat shock protein70/induced heat shock protein68 (HSC70/HSP68) and proliferating cell nuclear antigens (PCNA) during rat liver regeneration were analyzed. The results showed that their activities and con-tents were increased 4 hours after the lobus sinister was resected at the first time partial hepatectomy (PH).The activity of the ACP and content of PCNA were negatively relat-ed with AKP and HSC70/HSP68 4 hours after the 2nd and 3rd time PH respectively,when the centralis and dexter were resected,and that the PCNA expression was positive with the number of sequence of CPH.Based on changes of the distribution,content and activity of ACP and AKP in the rat liver by CPH,it was inferred that the AKP located in cell membrane regulated the cell division and differentiation through activiating ACP in cytoplasm,and that PCNA and HSC70/HSP68 may play an important role in liver regeneration by participating in synthesis,transport,selective degradation of proteins and DNA replication.

The chicken embryos incubated for 8?12 days were trypsinized to dissociate the chicken embryonic fibroblasts (CEF). CEF were maintained in DMEM containing 4.5 g/L glucose,10% newborn calf serum(NBS) as the feeder layer for culturing chick-en ES cells after treated with 10 mg/L Mitomycin C. The step down test and morris water maze test were applied to examine the learning and acquisition ability of the transgenic mice of Alzhimer’s disease. In step down test,the APP695 transgenic mice at the age of 3 months showed siginificant difference in counts of errors and latency time as comparing with those of the control group (
P
<0.01). In water maze test,9 months old APP695 and APP751 transgenic mice showed great difference in total counts and latency time as compared with those of the control group. It was suggested that the transgenic model’s behavior was conformed to the behavior of the early stage of human Alzhimer’s disease.

=28). The atherosclerotic lesions were developed by inducing endothelial denudation with bal-loon catheter and being given high dosage of cholesterol diet at the same time. The artery blood were sampled and iliac arteries were removed at 1,2,4,8 weeks after en-dothelial injury respectively. The results showed that the level of TC,TG,LDL-C and intimal thickness,intima/media(I/M)increased continually with the time.Coefficient of linear correlation between intimal thickness and TC、TG、LDL-C were 0. 8981、0. 8793 and 0. 8682 (

<0.01) respectively. The results suggested that clinic character of the process of atherosclerosis in the animal model was similar to that in human.

To establish an allergic rhinitis model induced by

=20) and an unsen-sitized control group(

=10).Sensitized groups were immunized intraperitoneally and intracutaneously with
Ascaris Imnbricoides
allergen for 4 weeks,followed by intranasal administration. Unsensitized control group were treated with 0.9% NaCl instead of As-caris lumbricoides allergen. As a results,the symptoms of allergic rhinitis,such as sneezing,itching and rhinorrhea were observed in 18 guinea pigs of sensitized groups with significant increase in the number of eosinophils and mast cell in the epithelium after 30 min. The results suggested that the successfully established animal model of aller-gic rhinitis may be used as a tool for the study and treatment of allergic rhinitis,especial-ly induced by
Ascaris lumbricoides
allergen.

To study the placental and fetal heart function,cardiopulmonary bypass was established in fetal lambs and the different methods of cannulation were applied.The results showed that the direct pulmonary artery cannulating was more reliable than pulmonary artery cannula ting through right ventricular outflow tract.Besides,the warm-ing of the fetal lamb,avoiding of uterine contraction,umbilical cord twist and bleeding during operation should be also emphasized.

To investigate the influence of magnetized water on the experimental hyperlipemia and atherosclerosis (AS) in rabbits,the rabbits were fed with hypercholesterol food and magnetized water for 90 days,the blood lipid level was detected and the aortic intimate were examined in all rabbits.The results showed that the plasma cholesterol,low-density lipoprotein and triglyceride of the animals feeding with magnetized water were significantly lower than those of the control group,whereas the area of fatty steak and atheromatous plaque were less. It can be concluded that the magnetized water plays role in prevention and treatment of the experimental AS in rabbits.

The methods of establishment of tumor metastasis model in nude mice were reviewed.When screening tumor metastasis cell line,both vivo and vitro methods were always carried out at the same time,one was to reinoculate cancer cells until acquired high metastasis cell line,the other was to get cancer cell subline which had metastasis ability by culturing in vitro. The author also pointed some issues which worth for further study.
