    Establishment of Mouse Model of P. acnes-mediated Liver Granuloma and Mechanism Investigation

    Objective To establish a mouse model of liver granuloma and investigate its mechanism. Methods C57BL/6J mice were injected Propionibactenium acnes (P.acnes) via tail vein. After injection, number of F4/80-B220-CD 11c+ cells and level of ALT were detected at time points of 1,6 h,and 1,2,3,5 and 7 d. On the 7th day after injection, pathological changes of livers between treated mice and untreated normal mice were compared. Nad’ve mice were injected with the sorted F4/80-B220-CD1lc+ cells from peripheral blood of P.acnes-treated mice to establish an adoptive transfer group. Results Number of F4/80-B220-CD11c+ cells and level of ALT upgrade obviously after F4/80-B220-CD11c+ cell inflision as well as P.acnes injection. Pathologic examination showed that granulomas were formed in the livers either in P.acnes or in F4/80-B220-CD1lc+ cell induced mice, accompanied with inflammatory cells infiltration.Conclusion P.acnes can induce liver granuloma of mice successfully and F4/80-B220- CDllc+ cell play an important role in liver granuloma development.
    Establishing Human Prostate Cancer Models from Different Cells and Its Metastatic Character

    Objective To compare the biological character between the model of human prostate cancer cell DU 145 and 22Rvl utilizing ectopic and orthotopic implantation in nude mice. Methods The human prostate cancer cell DU 145 and 22Rvl were inoculated under orthotopic or ectopic environments separately in nude mice, the mice were were sacrificed at the agonal stage. The tumorigenicity and lymph node metastasis were determined by gross and histological examinations. Results The 22Rvl cells were equally tumorigenic with 10/10 frequency in the prostate and subcutis, they exhibited highly metastasis potentiality with 9/10 lymph node metastasis after orthotopic implantation in nude mice for 40 days, whereas no lymph node metastasis after ectopic implantation.The DU145 cells were equal tumorigenic with 10/10 frequency in the prostate and subcutis, they exhibited lowly metastasis potentiality with 1/10 lymph node metastasis after orthotopic implantation in nude mice, and no lymph node metastasis after ectopic implantation.Conclusion The lymph node metastasis model established by orthotopic implantation in nude mice is successful, and it may become a valuable model for the study of metastasis mechanism and experimental therapy of prostate cancer.

    Objective To establish a mouse model of CD8+T cell-mediated acute graft versus host disease(GVHD). Methods Female C3H.SW(H-2Db, CD45.2+)mice was used as donors and B6/SJL(H-2Db, CD45.1+)mice as recipients, both of them are MHC-identical but miHA-mismatched. Before transplantation, the B6 recipients received 137γ ray (9.5 Gy) TBI administered in three treatments from a 137Cs source, then the T cell-depleted bone marrow (T-BM) cells and CD8+T cells from C3H.SW donors were injected immediately via tail vein. Results Two of the recipients injected with C3H.SW TBM and C3H.SW CD8+T cells died 28 days and one died 70 days after transplantation. The recipients injected only with C3H.SW T-BM were still alive within 70 days after transplantation. B6 mice without transplantation died within 6-10 days after irradiation. Flow cytometric analysis showed that mononuclear cells from the bone marrow, spleens and livers of B6 mice derived from the donor, and part of mononuclear cells from the spleens and livers was CD45.2+,CD8+T cell. Pathologic examination of the liver showed that the accumulation of mononuclear cells was in the portal areas of the liver. The structure of skin was destroyed. Conclusions The miHA-mismatched mouse model was CD8+T cell-mediated acute GVHD model. The MHC-identical, multiple miHA mismatched mouse model is analogous to human clinical miHA-mismatched bone marrow transplantation, which can be used as a good tool for study of pathogenesis and prophylaxis of GVHD.

    Objective Comparative the results of PCR and serology methods for detecting of three different pathogenes. Methods The nucleic acid of Toxoplasma gondii、Brucella and Rotavirus extracted from the liver of ICR mice、stool speciments and vagina specimen of Beagle、stool specimen of Macaca mulata respectively was amplified by NT-PCR、PCR and RT-PCR. The serum antibody of Toxoplasma gondiiBrucella and Rotavirus from ICR mice、Beagle and Macaca mulata respectively was detected by serology methods. Results The results of NT-PCR、PCR、RT-PCR were accorded with serology methods.Conclusion The developed NT-PCR、PCR、RT-PCR assay was a rapid, sensitive and specific method, it can be used in monitoring pathogen.

    Objective To investigate the expression of p21WAF-1 in brain tissue of the acute brain aging rat models, which will provide theory for brain-aging mechanism and clinic prevention and therapy. Methods The rat acute brain-aging model was established by60Co radiation, and 5-monthly SD rats as the control group. The learning and memory of rats ability was determined by Y type labyrinth, and the antioxidant and lipid peroxidative injury of rats were measured by SOD and MDA in blood. With the in situ hybridization, the expression of p21WAF-1 in hippocampal neuron was measured. Results The learning and memory ability of brain aging rat was decreased with Y type labyrinth. The blood’s SOD in brain-aging model was significantly lower than that of the control, the blood’s MDA in brain-aging model was markedly higher than that of the control. Over expression of p21WAF-1 was found in hippocampus, cortex, in hypothalamus and corpus striatum secondly, and in cerebellum est, the expression of p21WAF-1 were found in cone cells of cortex and hippocampus, in purkinje cells of cerebellum;the expression of p21WAF-1 were negative in cerebellum of control group, and the expressions of p21WAF-1 were found in other different brain regions in control group, but lower and weaker. Conclusions The simple and easy acute brain-aging model was established.Over expression of p21WAF-1 may maintain the character of senile neuron.

    Objective To investigate the changes of collagen and MMPs after β irradiation mimicked by electrons-ray. Methods An animal model of β irradiation injury was established by using linear accelerator electron radiation. The expressions of collagen and MMP1, MMP2, MMP9 were measured. Results After electrons irradiation, the expression and the structure of collagen were changed. MMP1 was increased. The expression of MMP2 and MMP9 were also changed. At first they were decreased and then increased. Conclusion Difficult healing of the wound caused by β irradiation may have a relationship with the collagen change, which may be connected with the activity change of MMP1. At the same time MMP2 and MMP9 can help the wound healing.

    Objective To observe the effect of Guanxinning tablet on hemorrheology and correlative biochemical indexes in aged rats. Methods Fifty 15-month-old male SD rats were randomly divided into 5 groups including a model control group, a positive control group and low (0.3g·kg-1), middle (0.6g·kg-1) and high (1.2 g·kg-1) dose groups of Guanxinning tablet. An additional ten 3-month-old rats were used as the normal control group. Determine the hemorrheology and the biochemical indexes of glucose (GLU), total cholesterol (TC), triglyeride (TG), superoxide dismutase (SOD) and malondialdehyde (MDA) in blood serum of rats after 15 days. Results Guanxinning tablet with low, middle and high dose had significant depressed effect on the level of blood viscosity, plasma viscosity, erythrocyte rigidity index, and significant elevated effect on the activity of SOD and depressed effect on the content of MDA in serum of aged rats. Guanxinning tablet with high dose had significant depressed effect on the level of TC and GLU. Conclusion Guanxinning tablet had significant amendatory effect on the abnormity of blood viscosity and erythrocyte rheological characteristic in aged rats, and contributed to inhibit the peroxidation of free radical, protect the activity of anti-oxidative enzyme, boost elimination ability for free radical. In a word, Guanxinning tablet had the characteristic of promoting blood circulation by removing blood stasis.

    Objective To investigate the effects of H2 receptors in the locus ceruleus (LC) on carotid sinus reflex (CSR) resetting resulted from the stress. Methods Male Sprague-Dawley rats were randomly divided into two groups: unstressed (n=8) and stressed group (n=9). The latter was subjected to unavoidable electric foot-shock twice daily for a week, each session of foot-shock lasted 2 hours. The left and right carotid sinus regions were isolated from the systemic circulation in all animals anesthetized with pentobarbital sodium. The intracarotid sinus pressure (ISP) was altered in a stepwise manner in vivo. ISP-mean arterial pressure (MAP) relationship curve and its characteristic parameters were constructed by fitting to the logistic function with five parameters. We observed the changes in CSR performance induced by stress and the effects of microinjection with H2 receptor selective antagonist, cimetidine (CIM), into the LC on the responses of CSR to stress. Results Microinjection of CIM (1.5 /μg, in 1 μl) significantly shifted posterior half range of ISP-MAP relationship curve downwards (P<0.05) and obviously decreased the value of the reflex parameters such as threshold pressure, saturation pressure and ISP at maximum gain (P<0.05) in the stressed rats. Microinjection with the same dose of CIM into the LC in the unstressed rats did not change CSR performance significantly (P>0.05). The changes in the stressed CSR performance resulted from the microinjection of CIM into the LC did not recover the levels of unstressed CSR after injection. Conclusion H2 receptors in the LC is involved in the stress-induced CSR resetting, and at the same time, the effects of stress on CSR also have other mechanism.

    Sperm cryopreservation is a well-established technique for most mammals. Whereas, a successful cryopreservation protocol of rodent sperm remains a scientific challenge. Especially for rat, its long tail of sperm makes its sperm cryopreservation very hard. Accordingly,ICSI of rat is not done better than mouse. There are not any reports on rat sperm cryopreservation and Intracytoplasmic sperm injection(ICSI) in China, and the researches were not done widely abroard .The artical made a simple review on the advances of sperm cryopreservation and Intracytoplasmic sperm injection(ICSI) in rat.
