不同的人癌裸鼠移植瘤模型中磷脂酶A2的表达

Expression of Phospholipase A2 in Different Xenografted Tumor Models of Nude Mice

摘要: 目的 观察磷脂酶A2(PLA2)在不同的人癌裸小鼠移植瘤模型中的蛋白表达及特征。方法 分别采用肺癌、鼻咽癌、肝癌、胰腺癌、胃癌、结肠癌、食管癌和卵巢癌肿瘤细胞株构建裸小鼠皮下移植瘤模型,待肿瘤生长至瘤体积达1 cm3后,采用酶联免疫吸附(ELISA)法检测肿瘤组织、正常组织及血清中4种主要类型PLA2(cPLA2、sPLA2、iPLA2和lp-PLA2)的蛋白表达,并以正常裸小鼠血清作对照。结果 在呼吸系统、消化系统和生殖系统的8种人肿瘤细胞裸小鼠移植瘤模型中,肿瘤组织cPLA2、sPLA2、lp-PLA2的蛋白表达均显著或极显著高于正常组织(P<0.05,P<0.01),肿瘤组织iPLA2的蛋白表达均高于正常组织,除人结直肠癌细胞HT-29移植瘤模型外,均呈现极显著性差异(P<0.01)。8种移植瘤模型中,荷瘤小鼠血清中4种类型PLA2的蛋白含量均极显著高于正常小鼠(P<0.01)。结论 4种类型PLA2在呼吸系统、消化系统和生殖系统不同的裸小鼠移植瘤模型的肿瘤组织和血清中均呈现高表达,无肿瘤类型特异性,表明磷脂酶A2可能参与肿瘤的发生、发展。

Abstract: Objective To observe the expression of phospholipase A2 (PLA2) in different human cancer xenograft models in nude mice. Methods Different human cancer xenograft models, including hepatocarcinoma, pancreatic cancer, gastric cancer, colon cancer, esophageal cancer, lung cancer, nasopharyngeal carcinoma, ovarian cancer, were established by tumor cell lines in nude mice. After the tumors grow to a volume of 1 cm3, the protein expression of four types of PLA2 enzymes in the tumor tissues, normal tissues, and serum were detected by ELISA, which in the serum of normal nude mice were tested as control. Results In the nude mice xenograft models of respiratory, digestive and reproductive system, the protein expressions of cPLA2, sPLA2 and lp-PLA2 in tumor tissues were significantly higher than those in normal tissues (P<0.05, P<0.01). Similarly, the protein expression of iPLA2 in tumor tissues was higher than that in normal tissues, except for human colorectal cancer cell line HT-29, which showed significant difference (P<0.01). Moreover, in the eight xenograft models, the protein content of four types of PLA2 in serum of tumor-bearing mice were obviously higher than those of normal nude mice (P<0.01). Conclusions Expression of four types of PLA2 in tumor tissues and serum were both up-regulated significantly in all the xenografted models, including respiratory cancer, digestive system cancer and reproductive system cancer, which did not limit to specific cancer type. The results inferred a key role for the PLA2 in the development of xenografted tumors in nude mice.
