巴马小型猪动脉粥样硬化模型相关炎症因子的表达

Expression of Inflammatory Factors in Atherosclerosis Model of Bama Mini-pig

摘要: 目的 观测高脂诱导巴马小型猪动脉粥样硬化(AS)模型腹主动脉和冠状动脉相关炎症因子表达情况。方法 取4~5月龄巴马小型猪随机分成正常对照组、模型组,每组各5只。正常对照组饲喂基础饲料,模型组均饲喂高脂饲料,连续饲喂 24周,取腹主动脉和冠状动脉进行油红“O”脂肪染色和常规HE染色,并进行免疫组化观测单核细胞趋化蛋白-1(MCP-1)、核因子-κB p65(NF-κB p65)、细胞间黏附分子1(ICAM-1)和血管细胞黏附分子1(VCAM-1)蛋白表达水平。结果 油红O染色显示模型组腹主动脉可见明显脂纹沉积; HE染色显示模型组腹主动脉内膜明显增厚,为AS病灶III型,冠状动脉可见动脉粥样硬化斑块形成,为AS病灶IV型; 免疫组化观察结果,与正常对照组比较,模型组腹主动脉MCP-1、NF-κB p65和VCAM-1蛋白表达水平表达显著增加(P<0.05),冠状动脉MCP-1、NF-κB p65、ICAM-1和VCAM-1阳性表达均显著增加(P<0.05)。结论 巴马小型猪AS模型腹主动脉和冠状动脉病变符合临床AS病变特征,其MCP-1、NF-κB p65、ICAM-1、VCAM-1蛋白表达均不同程度升高,与临床AS炎症反应和发生机制相符。

Abstract: Objective To observe the expression of MCP-1, NF-κB p65, ICAM-1and VCAM-1 protein in the abdominal aorta and coronary of the atherosclerosis (AS) model of Bama mini-pig induced by high fat diet. Method Bama mini-pigs which were 4-5 months of age are divided into normal control group which are fed basal diet for 24 weeks normal and model group which are fed high fat diet for 24 weeks, each group included 5 ones. Then the abdominal aorta and coronary of these mini-pigs were taken to conduct oil red “O” fat stain and routine HE staining. The MCP-1, NF-κB p65, ICAM-1 and VCAM-1 protein expression were observed by Immunohistochemistry staining. Result The result of oil red “O” staining shows that arty streak deposition was visibly on the abdominal aortic in model group and HE staining showed that in model group, abdominal aorta obviously thickening which is type IV of AS lesions and the formation of atherosclerotic plaque in coronary artery. The result of Immunohistochemistry staining was that, to contrast with normal group, the MCP-1, NF-κB p65, ICAM-1and VCAM-1 protein expression in abdominal aorta of model group are significantly higher(P<0.05); the MCP-1, NF-κB p65, ICAM-1and VCAM-1 protein expression in coronary of model group are significantly higher(P<0.05) too. Conclusion Abdominal aorta and coronary artery lesions of the AS model of Bama miniature pig consistent with clinical pathological characteristics of human AS. The MCP-1, NF-κB p65, ICAM-1and VCAM-1 protein expression are rise in different degree, which consistent with the clinical AS inflammatory reaction and mechanism.
